Modification of tomato traits and phytohormonal signaling using Target-AID for efficient nucleotide substitution

Recently, scientists at the University of Tsukuba have developed a new CRISPR / Cas9-based genome editing tool called Target-AID, which induces activation of Target. They confirmed and expanded the applicability of the Target-AID genome editing system by targeting SlDELLA and SlETR with the model tomato variety “Micro-Tom” and commercial tomato varieties.

After confirming that the Target-AID system induces nucleotide substitutions, we isolated mutants showing high GA sensitivity in both “Micro-Tom” and commercial varieties. Nucleotide substitution is targeting with a single sgRNA and multiple sgRNAs within a multi-single vector. In addition, the Target-AID system also generated Indel’s Target-AID-specific patterns with different editing positions than the Cas9 vector.

“We show that the Target-AID system can efficiently edit sgRNA targeting sites and facilitate case studies on the application of base editing techniques to translational research aimed at commercializing genome-edited crops,” Ezura said. The professor says. These results show that the Target-AID genome editing system is a promising tool for molecular breeding of tomato crops, and the scientific and scientific use of Target-AID in combination with other base editing systems. It highlights the agricultural potential.
