The development of the field of metagenomics, the study of genetic material from environmental samples, has revolutionized the way we observe and discover new species. Many bacteria cannot be cultivated independently in the laboratory. This may be due to the inadequate medium in which they grow, or because these bacteria only propagate in multiple communities (such as many in the gut). It may also be because it can only grow in relation to another larger organism. .. The group of bacteria that belong to the final category is Candidatus phylum Dependentiae. So far, little is known about this group, as only three strains belonging to this group have been isolated. However, recent studies have shown that Announcement of microbiology resourcesProfessor Masaharu Takemura of Tokyo University of Science (TUS) succeeded in isolating such a fourth strain, Noda2021.
“Initially, we aimed to sample the Risoukai Park on the Noda Campus of Tokyo University of Science and use Vermamoebavermiformis, a general laboratory host, to screen for giant viruses. I happened to discover this rare bacterium that also infects me, “says Dr. Takemura.
To isolate the new strain, Dr. Takemura first cultivated a sample taken from a pond in Rizokai Park and added it to the culture of Verma Moeva. After growing Verma Moeva for several days, he extracted Noda 2021 from it and analyzed its genetic material.
“The Noda 2021 strain was found to be composed of 1,222,284 base pairs with a guanine and cytosine (GC) content of approximately 38.3% and 1,287 genes. Next, a 16S rRNA molecular phylogenetic analysis of this strain was performed. By the way, other Candidatusphylum Dependentiae stock Dr. Takemura described it as the “piriformis muscle” that had been isolated so far, and when he examined infected Vermamoeva cells under an electron microscope, he discovered that Noda2021 may show a connected cell structure within the host cell. Did.
“This finding is evidence that the ponds on the Noda campus are microbiologically diverse and ecologically stimulating,” says Dr. Takemura. This is the first time such a strain has been isolated in Japan.
The isolation of this new strain of Candidatus phylum Dependentiae must further enhance our understanding of this curious bacterial population. According to Dr. Takemura, “Since this bacterium is located in the border area between giant viruses and microorganisms, we hope to provide some useful and unique information about the origin and ecological position of both groups. doing.”
Indeed, the Noda Campus of Tokyo University of Science seems to have many hidden treasures for emerging microbiologists. We cannot wait for the next discovery, whether by chance or not.
