Human primordial germ cells (PGCs) are the early progenitor cells of eggs (oocytes) and sperms needed to maintain human survival and reproduction. Medical researchers are using models of these cells, called human primordial germ cell-like cells (PGCLCs), to study their function and development.

Human PGCLC has been developed at several research centers, including the Cancer Research Center at Massachusetts General Hospital (MGH) and a laboratory led by Satoshi Shioda (MD, Ph.D.) at Harvard Medical School.

Natural PGCs are only present in embryos. The model was created to overcome the ethical and technical barriers surrounding the use of human embryonic tissue in experiments.

“My main interest is to use PGCLC for toxicological studies to understand how exposure to chemicals is carried out. Prescription drugs May affect Human reproduction“Shioda says.

He and his colleagues use cells to show how exposure of women to chemicals such as general anesthetics introduces hereditary changes and passes them on to future generations without causing DNA mutations. I am researching in the laboratory whether it can be done. This process is known as epigenetic inheritance.

They are also using this model to investigate what the PGC will look like Testicular cancerThe most common malignancies in boys and young men.

The catch is that hPGCLC cannot survive long in the laboratory and quickly loses germ cell-like features unless carefully and carefully cared for with the use of serum or additional chemicals. And even when these special cells survive and grow in laboratory dishes, they tend to drift away from germ cell-like identities and become other types of cells.

However, as Shioda et al. Explain in the journal. Stem cell report, They have developed a method for maintaining hPGCLC and its germ cell-like function in cell culture without the need for special treatment. The cells survive and continue to replicate for at least 5 months without losing their primitive germ cell-like function. ..

These cells were used to successfully generate hPGCLC with various genetic mutations associated with testicular cancer, and to support research in the prevention and treatment of cancer, human testicular cancer with defined genetic mutations. We are working on the creation of the first synthetic tumor model.
